چکیده

Introduction. The aim of the study was an analysis of pharmacokinetics and PK/PD parameters Cmax/MIC and AUC/MIC for ciprofloxacin in intensive care unit patients. Material and methods. Patients (n=3) received ciprofloxacin intravenously at 400 mg/ 8 h. Blood samples were collected on day 1st of the ciprofloxacin therapy. Plasma drug concentrations were measured by HPLC-UV method. Results. The mean maximum concentration after the first dose was Cmax = 3.26 (± 2.41) μg/ml. The mean Cmax/MIC and AUC/MIC ratio were: 10.48 (± 4.72) and 192.1 (± 134.8), respectively. Conclusions. Big inter-subject variability for PK/PD parameters in ICU patients requires systematic monitoring. AAnestezjologia i Ratownictwo 2010; 4: 409-413.

Optimal Aminoglycoside Therapy Following the Sepsis: How Much Is Too Much?

Severe sepsis and septic shock are major problems as the result of high rates morbidity andmortality in intensive care units (ICUs). In the presence of septic shock, each hour of delay inthe administration of effective antibiotics is associated with a measurable increase in mortality.Aminoglycosides are effective broad-spectrum antibiotics that are commonly used in ICUs forthe treatment of life...

Using Iron-Chelating Agents in Critically Ill Patients with Iron Overload. Fact or Fiction?

Recently, some evidence has shown that the failure of iron homeostasis may occur in critically ill patients and can lead to iron overload. Elevated ferritin levels as a body iron burden index in critically ill patients may be associated with depressed level of consciousness and greater mortality. However, the necessity of using iron-chelating agents in clinical situation is still unknown for th...

Monte Carlo simulations: maximizing antibiotic pharmacokinetic data to optimize clinical practice for critically ill patients.

Infections in critically ill patients continue to result in unacceptably high morbidity and mortality. Although few data exist for correlating antibiotic exposure with outcome, antibiotic dosing is likely to be highly important for maximizing resolution of infection in many patients. The practical and financial difficulties of performing pharmacokinetic (PK) studies in critically ill patients m...

The PK/PD index (CMAX/MIC) for ciprofloxacin in patients with cystic fibrosis.

In order to evaluate the efficacy of an antibacterial therapy, three basic PK/PD indexes were defined: the ratio between the maximum drug concentration obtained after a single dose and minimum inhibitory concentration MIC (CMAX/MIC), the ratio between the area under the curve of dependence between the drug concentration in blood and time within 24 hours to MIC (AUC24/MIC), and the time when the...
